Is it possible to have the keyword search feature already include certain terms in the search?

For example, if I want to make a keyword search with the keyword "dl" for a Google search that already includes the terms "site:mediafire.com" automatically.

Sure, just add that search term before the %s in the bookmark.
You probably can't do that if you add the keyword via the Manage Search engines, but it should work if you create a bookmark and assign a keyword to it.
