My email suddenly seems to have changed from UK English to American English (when typing favour, it was underlined as queried but when I deleted the "u", it was happy) How do I change the system to UK English?

Check the language setting and you might need to download new dictionaries. Thunderbird 68 uses a new style dictionary so if Thunderbird just updated, new dictionaries will be required.
