When I backup Windows Live Mail, I can go go to the backup copy and open my folders and emails and read them. Can I do this in Thunderbird?

Is that one profile folder I copied enough to bring my entire account back if something should happen?

Thunderbird does not use the EML format for mail so no you can not read from your backup.
