I've tried using message filters to automatically dispose of unwanted messages from specific people.

It works but the messages still appear in Trash.

You could set server settings to empty trash on exit. and expunge inbox

It means that unwanted mails will still be in trash until i exit Mozilla Thunderbird.

Is there anyway to completely block unwanted messages at all?

You cannot block message. Only email servers can block anything and you have no control over that. The suggestions you have so far is what you have to work with.

If you use a pop account.:

You have FiltaQuila addon. You leave message on popserver and download headers You keep all junk-addresses (that is full address) in an address-book (junkers)

Make this filter: If from is in your AB-JUNKERS then delete on pop-server and delete message.
