Check for Updates appears in the About Firefox dialog window when automatic updating for Firefox is turned off. No, when you click that button it checks and automatically downloads the update, if there is one available. No way to separate those as how it was in versions before Firefox 4.

Thanks - I didn't think there was but I figured I'd ask.

Yes it depends on how you have it set in Tools->Options (or Preferences) ->Advanced->Update. Having Automatic update off will make it so there is a "Check for Updates" button.
