Shut down Firefox. Using your file browser, open the Programs Folder on your computer.
Windows: C:\Program Files C:\Program Files (x86)
Right click on Firefox.exe and select Properties. Now select Compatibility. Turn Off Run This Program In Compatibility Mode, and Run This Program As An Administrator.
Do the same for any Firefox shortcuts you have. Now restart Firefox.
I am a Windows user, so this may be wrong, but I believe that on Mac, if you cannot use the built-in updater successfully, you can do a "manual" update by downloading the new version, quitting Firefox, "trashing" the old version (without removing your settings) and then installing the new version.

But you might wait for a Mac user to chime in.
Loved the thoroughness of the reply from FredMcD, except for the fact that i am on a Mac with OS X operating system. Any thoughts in that regard.
If there are problems with updating or with the permissions then best is to download the full version and trash the currently installed version to do a clean install of the new version.
Download a new copy of the Firefox application and save the disk image file to the desktop
- Firefox 40.0.3: https://www.mozilla.org/en-US/firefox/all/
- Trash the current Firefox application (open the Applications folder in the Finder and drag the Firefox application to the Trash) to do a clean (re)install
- Install the new version that you have downloaded

Your personal data is stored elsewhere in the Firefox profile folder, so you won't lose personal data like bookmarks when you uninstall and (re)install or update Firefox.
