Are you sure it is enabled, both in Tools > Options > Composition > Enable spell check as you type and in Options menu > Spell Check As You Type when composing a message?

Do you have a dictionary loaded and installed? In the Spell Check dialogue box, there is a drop-down at the bottom labelled "Languages". Or there is a drop-down button on the Toolbar button which lists languages.
