I am looking for a way to (quick) filter or search messages which have a specific "Reply to" header. For instance "Reply to: mymail@mail.com". Is there such a feature or add on available for Thunderbird?

Open a Classic Search (Ctrl+Shift+F) or filter window, click the drop-down menu that shows Subject, Customize..., type in Reply-To, Add, OK. Messages can now be searched or filtered on the Reply-To header.

This is exactly what I already tried (with different variations such as "reply to", "reply-to") but when I search for a string then no or only partial results are returned. The source code of the message contains a "Reply-To" header.

It works here (see picture). Make sure the custom header is Reply-To with no quotes.
