Thunderbird wants to do a new set up rather than opening my existing program

I've used TB for several years on a portable drive. In trying to open the program today, TB opens but thinks it's a new set up and wants me to set up all the parameters for the mail like it's a new account. I opened "profile" from Explorer and all my file folders show up, but the files indicate "empty". Help Please!! I have lot's of important messages I can't afford to lose.
