How to enable one manually previously disabled cookie?
I disabled 1 cookie of a particular site manually. I tried clearing the "Cookies and Site Data" afterwards. I am still unsure whether they are enabled now or not. Please assist.

Your first screenshot shows the exceptions for Enhanced Tracking Protection and not the cookie exceptions further down on the page.

Did you check the cookie exceptions in "Settings -> Privacy & Security" as that where such a denial is stored ?
See also the permissions icon the location bar and "Tools -> Page Info -> Permissions".
I checked them both, but I could not find anything.
