Was Thunderbird installed before you upgraded to Windows 10, or did you install Thunderbird after the upgrade?

Usually, there is no conflict between the Win10 install and Thunderbird, BUT... It is possible that something happened to make TB forget where your old profile is located. You may have to use the profile manager to 'point' TB to it. Try to verify that your profile still exists on your hard drive.
