After upgrading to FF 12 beta, a blank page is displayed instead of the home page and the bookmarks toolbar is missing.
When I go to View -> Toolbars -> and deselect Bookmarks Toolbar, and then again View -> Toolbars -> select Bookmarks Toolbar, the bookmarks appear.
Also, when starting up, a blank page instead of my homepage is shown. (Options -> General -> Show Homepage is selected)

You should take a look at the Knowledge Base article How to set the home page. There are steps in that article to reset your homepage preferences to default. I would try that first to clear out any strangeness. If that works, you can try setting your homepage again to your personal preference.
If that doesn't work you should look at the article How to fix preferences that won't save. I'd imagine that if your selection for showing the bookmarks toolbar isn't persistent then Firefox is probably having trouble writing to the pref.js file.
Start Firefox in Troubleshoot Firefox issues using Safe Mode to check if one of the extensions or if hardware acceleration is causing the problem (switch to the DEFAULT theme: Firefox/Tools > Add-ons > Appearance/Themes).
Don't make any changes on the Safe mode start window.
Now we are going to make another backup, with only the basics. BE SURE NOT TO OVERWRITE THE FIRST BACKUP YOU CREATED FROM ABOVE! Name this new one something like basic.pcv
