What mail provider are you using. 2FA is not something Thunderbird inherently supports. Thunderbird connects to the mail server often many times a minute, hence suggestions that you use app passwords and oauh authentication, but it depends on the provider as to what is the best approach.

For outlook.com Thunderbird will set itself up using oauth authentication, Microsoft's preferred authentication method, they refer to it a "Modern Authentication" for the account. You will go through an initial set of web pages served by outlook.com or office365.com where your 2FA will be utilized. After that there will be a token issued to Thunbird to use to connect you and your mail based on the permissions granted in the web page flow.

2FA is NOT an option for outlook.com.
